PTEN (bahasa Inggris: phosphatase and tensin homolog, PTEN) adalah berkas genetik pada kromosom 10 lokus 10q23.3 dengan ekspresietik berupa protein yang berfungsi sebagai enzim dengan nama PI3P. Mutasi genetik PTEN sering ditemukan pada banyak kasus kanker, sehingga gen ini sering disebut sebagai salah satu tumor suppressor.[5]
Tidak seperti umumnya protein fosfatase tirosina, protein ekspresi PTEN lebih banyak mengkatalitik reaksi defosforilasi pada substrat fosfoinositida dan melakukan fungsinya sebagai penghambat tumor dengan menekan lintasan AKT/PKB.
